Comments | Population: Caucasian. Characteristics: Transduced with a pQMSCV-pHSP70B-TRE-pCMVmin-EGFP-IRES-tTA construct. The heat-activated HSPA6/HSP70B' promoter drives bicistronic gene expression of EGFP and of the tetracycline-controlled transactivator (tTA), which is mediated by an internal ribosomal entry site (IRES). Then, the tTA protein binds to TRE and activates the cytomegalovirus minimum promoter (pCMVmin) upstream of EGFP, which in turn induces further expression of the EGFP and tTA genes, providing a transcriptional positive feedback loop (PubMed=30962099).
