Abstract | The author established in vitro cell lines derived from two glioblastomas of the occipital and occipital regions of the human cerebral palsy. It has survived for 5 years and passed over 500 generations. Methods and Results 1. Establishment of cell lines: 1. Source of material: TJ899 cell line was obtained from a surgically removed specimen of a male patient with occipital glioma. The TJ905 in vitro cell line was obtained from a male TOP glioma patient. Both were diagnosed as glioblastoma multiforme by histopathology. 2. Construction: After aseptic conditions, the tissue was rinsed with Hanks' solution to remove blood and visible electrocautery and necrotic tissue. Mechanically fragmented, inoculate the tissue block in a 25cm~2 flask, add Waymouth's MAB 87/3 (Gibco Lab, USA) containing 15% fetal bovine serum (FCS), incubate at 37 Celsius, 5% CO2 72 hour. Most tissue blocks have different degrees of cell migration, and the cell body is small. After 1 week, most of the culture flask walls are full of cells. |
