Pathway activation results in the degradation of the target protein. The degraded protein is detected by adding to the cell lysis buffer another beta-galactosidase fragment (Enzyme Acceptor=EA). Complementation of the ED and EA enzyme fragments result into a catalytically active enzyme that generate a chemiluminescent signal.
